Description

Im Zentrum dieses Verfahrens steht die Entwicklung einer innovativen Plattform zur rasterkraftmikroskopischen Analyse biologischer Proben auf molekularer und zellulärer Ebene, sowohl unter Standard-Biosicherheitsbedingungen (S2) als auch unter hohen Sicherheitsauflagen (S3). Ziel ist es, dynamische Prozesse wie Protein-Protein-Interaktionen, Zell-Zell-Adhäsion, Rezeptorbindung oder die mechanische Entfaltung von Molekülen in realitätsnahen Umgebungen zu untersuchen - also ohne Fixierung, Dehydrierung oder Kryokonservierung. Die gewonnenen Daten sollen helfen, grundlegende biophysikalische Mechanismen z. B. bei Infektionsprozessen, Antibiotikainteraktionen oder Membranmodifikationen zu verstehen. Durch die Kombination von Rasterkraftmikroskopie (AFM) mit korrelativer Fluoreszenzbildgebung wird eine Brücke zwischen Struktur, Dynamik und Funktion geschlagen. Die neuartige Konfiguration erlaubt es dabei erstmals, solche Experimente unter S3-Bedingungen durchzuführen, was weltweit einzigartig ist. Die Universität Hamburg verfolgt mit dieser Infrastruktur das Ziel, eine führende Rolle im Bereich hochsicherheitskompatibler Einzelmolekül-Mikroskopie einzunehmen. Die Geräte werden durch kooperierende Gruppen innerhalb des CSSB (Centre for Structural Systems Biology) betrieben und genutzt. Gegenstand dieses Verfahrens ist die Lieferung und Installation spezieller Erweiterungskomponenten zur Einrichtung von zwei modularen Rasterkraftmikroskopsystemen (AFMs), basierend auf einer bestehenden JPK NanoWizard-Plattform (Bruker). Ziel ist der parallele Einsatz eines Systems im S3-Labor und eines Systems im S2-Labor am CSSB, im Wesentlichen bestehend aus: ▪ 1. Erweiterung zum Aufbau und Installation eines Wide-Range-AFM-Systems (S3-Labor), ▪ 2. Erweiterung zum Aufbau und Installation eines High-Speed-AFM-Systems (S2-Labor), ▪ 3. Ergänzung durch eine geschlossene Messkammer (S2-kompatibel).
