Description

Das IBMM möchte ein fabrikneues cryo-konfokales Fluoreszenzmikroskop (cryo-kFM) beschaffen, welches in einen cryo-CLEM Workflow für zelluläre cryo-ET und in situ Strukturbiologie integriert werden soll. Das cryo-kFM soll mit mindestens 4 Fluoreszenzkanälen (Lasern) ausgestattet sein sowie über ein hochsensitives, hochauflösendes Detektorsystem verfügen für die Konfokalmikroskopie. Das Mikroskopsystem soll es zudem erlauben, zwischen Konfokal- und Widefield-Fluoreszenzmikroskopie zu wechseln, indem es mit entsprechenden Lichtquellen, Filtern und Kamera ausgestattet ist. Zudem soll es mit mindestens einem cryo-kompatiblen Luftobjektiv mit möglichst hoher numerischer Apertur und einer mindestens 50-fachen Vergrösserung ausgestattet sein. Weiter soll das cryo-kFM über eine cryo-Stage verfügen, welche es erlaubt die Proben (cryo-EM Grids mit vitrifizierten Zellen) bei < -150°C zu laden, abzubilden und auszuladen, ohne dass die Proben je eine höhere Temperatur ausgesetzt sind, und ohne dass die Proben durch kondensierende Luftfeuchtigkeit signifikant kontaminiert werden. Die cryo-Stage muss mit gängigen cryo-EM grids und sogenannten AutoGrids für die weiteren cryo-CLEM Schritte kompatibel sein. Die motorisierte cryo-Stage soll in die Benutzeroberfläche der Software zur Bedienung des cryo-kFM integriert sein. Diese Benutzeroberfläche soll möglichst einfach und effizient zu bedienen sein; das gesamte Mikroskopsystem soll über eine komplette, nutzungsbereite Hardware und Software verfügen. Besonders wichtig sind hohe Bildgebungsqualität, Robustheit bei der Bergung der abgebildeten Proben ohne Qualitätsverlust für die nächsten cryo-CLEM Schritte (cryo-FIB milling und cryo-ET), sowie Benutzerfreundlichkeit.
